Abstract

ABSTRACT Background : The content of bioactive compounds as metabolites in plants that can be developed into medicinal raw materials. One of them is the nutmeg plant (Myristica fragfrans) which is native to Indonesia. The nutmeg mace plant is known to contain chemical compounds, namely alkaloids, flavonoids, saponins, phenols and tannins, where these compounds are known to have bioactivity as antioxidants which function to ward off free radicals. Aim : To determine the antioxidant activity of the n-hexane fraction, chloroform fraction and ethyl acetate fraction of ethanol extract of nutmeg mace (Myristica fragrans). Method : Determination of the antioxidant activity of several nutmeg mace extract fractions using the DPPH free radical capture method (2,2-diphenyl-1 picrylhydrazyl) using UV-Vis spectrophotometry. Results : The results of the phytochemical screening on nutmeg mace were positive for containing alkaloids, flavonoids and saponins, but the results were negative for phenolic and tannin compounds. The results of thin layer chromatography using n-hexane eluent: ethyl acetate (8:2) obtained in the n hexane fraction Rf was 0.69, in the chloroform fraction Rf1 was 0.46, Rf2 was 0.84, and in the ethyl acetate fraction Rf1 is 0.53, Rf2 is 0.76. The results of the antioxidant activity test of nutmeg mace (Myristica fragrans) showed that the ethanol extract sample had an IC50 value of 51.52 in the strong category, the n hexane fraction had an IC50 value in the strong category, the chloroform fraction had an IC50 value of 22.17 in the very strong category and the ethyl acetate fraction has an IC50 value of 129.89 in the medium strong category. Conclusion : The 96% ethanol extract of nutmeg mace (Myristica fragrans) contains secondary metabolite compounds of alkaloids, flavonoids, saponins, phenols and tannins so it has antioxidant activity in the strong and very strong categories with an IC50 value from the ethanol extract of 51.52 ppm, in fraction n -hexane was 71.57 ppm, in the chloroform fraction it was 22.17 and in the ethyl acetate fraction it was 129.89 ppm.
