Abstract
ABSTRAK Latar Belakang: Diabetes melitus (DM) merupakan penyakit degeneratif yang memiliki prevalensi kematian tertinggi yaitu 50% diantaranya akibat komplikasi DM. Jantung menjadi organ yang terdampak besar komplikasi DM akibat dari peningkatan produksi radikal bebas dan stress oksidatif yang dimediasi oleh kenaikan kadar gula darah atau hiperglikemik sehingga mengaktivasi jalur apoptosis dan nekrosis histologi sel miosit jantung. Tujuan: Untuk mengetahui pengaruh ekstrak etanol daging buah pala (Myristica fragrans Houtt) pada histopatologi jantung tikus wistar (Rattus novergicus) hiperglikemik. Metode: Hewan uji diinduksi pakan diet gula selama 7 hari dan dilanjutkan pada pemberian bahan uji masing-masing selama 1 minggu terhadap 25 ekor tikus yang dibagi menjadi 5 ekor dalam setiap kelompok perlakuan (kelompok 1 diberikan Na-CMC 0,5%, kelompok 2 diberikan metformin 500 mg, kelompok 3 diberikan EDBP 100 mg/KgBB, kelompok 4 diberikan EDBP 200 mg/KgBB, dan kelompok 5 diberikan EDBP 300 mg/KgBB). Parameter yang diamati meliputi perubahan bobot badan, kadar gula darah, bobot organ jantung, dan histopatologi jantung. Data kemudian dianalisis dengan uji One Way Anova dan dilanjutkan pada uji Post Hoc Mann-Whitney dengan taraf kepercayaan 95% (p<0,05). Hasil: Seluruh kelompok perlakuan ekstrak etanol daging buah pala (EDBP) menunjukkan perbaikan pada histopatologi jantung tikus yang berbeda bermakna terhadap kelompok kontrol (p<0,05) dan tidak berbeda bermakna terhadap kelompok metformin sebagai pembanding (p>0,05). Simpulan: Pemberian ekstrak etanol daging buah pala dapat memperbaiki kerusakan histopatologi jantung tikus wistar hiperglikemik pada dosis 100 mg/KgBB, 200 mg/KgBB, dan 300 mg/KgBB. Disi1, Ermalyanti Fiskia1 1Pharmacy Study Program, Faculty of Medicine, Khairun University ABSTRACT Introdution: Diabetes mellitus (DM) is a degenerative disease with the highest prevalence of death, with 50% of deaths due to DM complications. The heart is an organ greatly affected by DM complications due to increased production of free radicals and oxidative stress mediated by increased blood sugar levels or hyperglycemia, which activates the apoptosis pathway and histological necrosis of cardiac myocyte cells. Aim: To determine the effect of ethanol extract of nutmeg pulp (Myristica fragrans Houtt) on the histopathology of the heart of hyperglycemic Wistar rats (Rattus novergicus). Methods: The test animals were fed a sugar diet for 7 days and continued to administer the test material for 1 week each to 25 rats divided into 5 rats in each treatment group (group 1 was given Na-CMC 0.5%, group 2 was given metformin 500 mg, group 3 was given EDBP 100 mg/KgBW, group 4 was given EDBP 200 mg/KgBW, and group 5 was given EDBP 300 mg/KgBW). The parameters observed included changes in body weight, blood sugar levels, heart organ weight, and heart histopathology. The data was then analyzed using the One Way Anova test and continued with the Mann-Whitney Post Hoc test with a confidence level of 95% (p<0.05). Results: All groups treated with ethanol extract of nutmeg pulp (EDBP) showed improvements in rat heart histopathology which were significantly different from the control group (p<0.05) and not significantly different from the metformin group as a comparison (p>0.05). Conclusion: Administration of ethanol extract of nutmeg pulp can improve the histopathological damage to the heart of hyperglycemic Wistar rats at effective doses of 100 mg/KgBW, 200 mg/KgBW, and 300 mg/KgBW.
